Overview
MAFFIN combines feature-quality selection, signal-intensity correction and maximal-density fold-change normalization. Its R implementation returns normalized intensities, normalization factors and variation summaries.
What can this tool do?
Common uses
- Normalize processed feature tables using the documented QC design.
Scientific fit
Primary function
Quality control and filtering
Data compatibility
Inputs
Feature or peak table, Sample metadata
Outputs
Feature or peak table, Human-readable report
Input formats
CSV/data frame with features, samples and QC-group annotations
Output formats
Normalized table, normalization factors and PRMAD summaries as an R list or CSV
Scope and considerations
Designed for
- Normalize metabolomics feature intensities using selected features and corrected signal responses.
Not designed for
- Confirming compound identity or replacing study design and scientific review.
Important requirements
- An appropriately annotated feature table; serial QC loading amounts for the signal-correction component.
- Feature-by-sample CSV/data frame with group annotations for blanks, QC and serial QC loading levels
Considerations
- Do not assume that missing serial-QC information can be reconstructed from ordinary study samples.
- Normalization can change biological contrasts; evaluate its assumptions for the study.
